ACC.25: ALPACA, lepodisiran keeps Lp(a) down for 540 days after a single dose (Steven Nissen)

Steven Nissen (Cleveland Clinic) on the phase 2 ALPACA trial (NCT05565742) of lepodisiran, Lilly's extended-duration siRNA against apo(a). The 400 mg dose lowered time-averaged Lp(a) by 93.9 percent from day 60 to 180; a single dose kept levels 53.4 percent below baseline at day 540 and two doses (baseline and day 180) held a 74.2 percent reduction at day 360, with no major safety signal. That durability is the argument for the infrequent dosing being tested in ACCLAIM-Lp(a). Recorded at ACC.25.
